Summation

Rb remains unphosphorylated in G1 phase, forming a stable complex with E2F:DP heterodimers, and thus actively represses the transcription of E2F-responsive genes. In late G1 or at the G1/S transition phase, Rb becomes phosphorylated by CycD:CDKA, resulting in the release of free and transcriptionally active E2F:DP heterodimers. E2F:DP then actively promotes transcription of genes whose products are required for DNA synthesis in S-phase (den Boer and Murray, 2000; Shen, 2001).
