Summation

Synthesis of lagging strand involves semidiscontinuous replication of DNA, which generates short DNA fragments called Okazaki fragments. Polymerase activity is similar on both leading and lagging strand but as POLD extends the Okazaki fragment, it encounters the 5' end of the downstream replication product and displaces it from the template strand, generating a flap. The flap is then cleaved to generate a nick, which is ligated to form the intact nascent strand.
