The POLD with the processive complex synthesize DNA in a continuous manner during leading strand synthesis. The largest subunit (POLD1) contains the polymerase and exonuclease activity, while the other subunits are involved in complex stabilization and interactions with PCNA. The 3' to 5' exonuclease proofreading ability of pol delta removes the misincorporated nucleotides (Schultz et al., 2007).
